Prepare 0.1 ug/ml Dye, 50 ml (adjust  volume for number of samples run)
 

• 5ul of  1mg/ml Dye
• 5 ml of 10x TEN
• 45 ml ddwater, preferably sterile

Mix in 50 ml Falcon tube and cover it with aluminum foil, to prevent photobleaching.
 

• Turn on Perkin-Elmer LS5B about 20 minutes prior to using to warm up.
• Set exitation and emmision slit widths to max (knobs on side of instrument).
• Set 360 um exitation, 460 um emission wavelengths.
• Blank machine:  Add 2 ml of dye soln. to cuvette and autozero by pressing autozero button.
• Add 20 ul of 10 nanogram/ul DNA std to blank to make 200 ng DNA standard.
• Type 200 on keyboard and press AutoConc button (hold).   Now readouts will approximate ng per sample.
• Add 2-5 ul of unknowns to each fresh 2 ml aliquot of Dye.
